APPLICATION,This method is applicable to the sampling and examination of brown rice for light filth particles to determine,compliance with Sections 4, 5 and 7 of the Food and Drugs Act. This method replaces ExFLB No 13 by Gardiner.,2. DEFINITION OF TERMS,A lot is defined as that amount (volume, weight, etc.) of the food which is produced, stored and/or shipped under,conditions as nearly uniform as possible, preferably designated by a common container code or marking, and, in,any event, consisting of not more than one variety, grade or type of product from a single identifiable source.,3. COLLECTION OF SAMPLES,3.1 Scrutinize the entire lot for live infestation. If live infestation is found, do not sample until after fumigation,or other effective treatment.,3.2 Obtain three sample units of at least 100 g each, selected at random from the lot, using appropriate,sampling equipment and containers. Three sample units constitute a sample.,3.3 Each sample unit must be kept separate and labelled 1, 2 and 3. Complete information respecting the lot,size, weight of individual containers, country of origin, exporter, importer, or domestic manufacturer, and,product and lot identification should be recorded and should accompany the sample.,4. MATERIALS AND SPECIAL EQUIPMENT,1) Balance,2) 2 L beaker with a lid (watch glass),3) Smooth magnetic stirring bar, teflon coated (1 x 5 cm),4) Stirrer hot plate,5) U.S. standard No. 140 sieve,6) Large glass funnel, approx. 14cm diam.,ExFLP-12,- 2 - April 1995,7) Wildman 2 L trap flask (AOAC 945.75 h(4), 15th ed.),8) Wash bottles,9) Suction filtration apparatus with Buchner or Hirsch funnel (5-7 cm perforated plate),10) Ruled filter paper. Filter paper should be larger than funnel plate (7-9 cm),11) Petri dishes to suit size of filter paper used,12) Stereoscopic microscope (30-70x),13) Concentrated HCl,14) Alcohol, 60% and 95%,15) Tween 80-60% alcohol solution. To 40 mL polysorbate 80 (ICI United States, Inc.) add 210 mL 60%,alcohol, and mix.,16) Tetrasodium EDTA alcohol solution. Dissolve 5 g Na4 EDTA in 100 ml H2O, add 150 mL of 95% alcohol,and mix.,17) Tween 80-60% alcohol and tetrasodium EDTA alcohol solution, 1:1 (v/v). Mix just before use.,18) Mineral oil. Paraffin oil, white, light 125/135 Saybolt Universal Viscosity (38°C), specific gravity, 0.840-,0.860 (24°C),19) Glycerol-95% alcohol mixture (1:1),5. PROCEDURE,The examination shall be carried out in accordance with the following instructions.,5.1 Analytical Units,5.1.1 In a random fashion from a sample unit, weigh 100 g. This 100 g constitutes an analytical unit.,5.1.2 Repeat step 5.1.1 for remaining two sample units.,5.2 Isolation - Light Filth,5.2.1 Transfer the analytical unit into a 2 L beaker. Add 1 L dilute HCl (50 mL conc. HCl and 950 mL water) and,a magnetic stirring bar. Cover with a watch glass.,5.2.2 Boil with stirring for 30 min.,5.2.3 Wet sieve contents of beaker through No. 140 sieve with hot (50-70oC) water until wash water is clear.,5.2.4 Quantitatively transfer contents of sieve to a 2 L trap flask with the aid of a large funnel (place the end of,trap rod up through the funnel stem) using 60% alcohol. Rinse sides of flask with 60% alcohol and bring,volume up to 800 mL.,5.2.5 Add 100 mL of a 1:1 Tween 80-60% alcohol and tetrasodium EDTA alcohol solution slowly down trap rod,holding top of stopper just below the top of the 60% alcohol level. Hand stir gently for 1 min.,5.2.6 Let stand for 4 min.,ExFLP-12,- 3 - April 1995,5.2.7 Add 50 mL mineral oil slowly down trap rod, holding top of stopper just above the top of the 60% alcohol,level.,5.2.8 Place on a hot hot plate and slowly bring stirrer to a maximum speed that does not produce audible or,visible splashing or a permanent emulsion. All of the material should be in suspension and a portion of,the stirring bar should be visible at the bottom of the vortex. Stir for 5 min.,5.2.9 Slowly add deaerated 60% alcohol down trap rod, maintaining top of stopper just above oil layer. When,stopper is almost fully raised, continue adding 60% alcohol until the oil layer passes the stopper and the,bottom of oil layer is approximately 1 cm above top of fully raised stopper.,5.2.10 Swirl stopper gently through lower portion of trap flask so as to suspend se……
